The fact that a single and different colony of E. coli was used for each lab testing was a minor and random SOE in this lab. Although the strain of E. coli never changed (DH5α), each E. Coli colony was sprung from one single bacterium, and will have slightly different characteristics as compared to other colonies. This is the reason why control plates are done and, because of it, comparisons of data were not done between different test days (a single colony was removed from the agar plate each day to grow, in order to acquire a pure culture). This SOE is unlikely to be significantly reduced at this point, as we have taken all the necessary precautions to obtain a pure culture for each test day, including aseptic technique and sterile equipment.
